Using the Spectrophotometer

The spectrophotometer is used to measure the density of cells in solution.
1. Turn on lamp; check to make sure it is at the correct wavelength.
2. Place 0.5 - 1 ml of sample in cuvette. Don't forget to prepare a blank, aka a sample with no cells (LB, H2O, etc.). Place the blank in the slot labeled "B".
3. Press "B", then "Measure Blank."
4. Measure samples by pressing the appropriate number. Re-zero the machine every few samples for accuracy.
